MBS now using pH gradient elution for Purification
Industry Standard Protocol for Antibody Purification
Antibody bound to Protein A resin eluted under acidic conditions
Neutralization
often does not result in complete restoration of antibody to natural state,
instead it results in:
- Population of fragmented antibody
- Low signal to noise ratio
- Poor efficiency
- Poor consistency in conjugations
- Suppressed antibody titer
MBS Protocol for Antibody Purification
Antibody bound to Protein A resin eluted by pH gradient to minimize exposure
to harsh conditions
Receptor vessel for eluted antibody prepared with neutralization
reagent for quick return to physiological pH
MBS has consistently achieved
95% or better purity as determined by SEC.
In addition to designing gentle purification protocols, we have also studied the properties of our protein A resin to determine maximum capacity for murine Ig as well as maximum flow rates. Data generated from this study has been incorporated into our protocols and has resulted in improved column utilization while minimizing processing times
