Ascites Antibody Production & Purification

In Vivo Monoclonal Antibody Production via Mouse Ascites (6-8 Weeks)
- All incoming cell lines require mycoplasma testing
- IFA primed mice are injected with a hybridoma cell line
- BALB/c, BALB/c hybrid, Nude and SCID mice are available; additional strains available upon request
- Ascites fluid develops over the next 10-14 days and is harvested over multiple taps
- Antibody concentration typically increases with each successive tap
- Ascites is pooled, centrifuged, and gauze filtered
- An average yield from BALB/c mice is 3-5mLs of ascites per mouse with an average antibody concentration of 3-5mg/mL
Production Services Include:
- Expansion and cryopreservation of a hybridoma cell line to 5 vials
- Aggregate filtration of the pooled ascites
- Testing of ascites for protein content, Ig concentration
- Storage of ascites at -20C
- GMP production report including copies of zonal electrophoresis and densitometry scans

Antibody Purification Services (1-2 weeks)
- Murine Monoclonal IgGs are purified by Protein A affinity chromatography, eluting by pH gradient to ensure gentlest conditions possible (read more)
- Concentration/dialysis to desired final buffer
- Testing of purified antibody for protein content, Ig concentration
- Sterile filtration of final product
- Addition of NaN3 upon request
Additional Services Include:
- Percent purity is determined by Size Exclusion Chromatography allowing differentiation between monomeric, dimerized, aggregated and fragmented antibody
- IsoElectric point determination to reveal isoforms or variants in the purified material
In Vitro Production
Roller Bottle Productions or Spinner Flasks
- Cultures are grown to a specific viability and then harvested for optimal cell or cell product preparation.
- Production sizes range from 1.0L to 100.0L
- GMP production report and copy of zonal electrophoresis
- Expansion and cryopreservation of cell lines to 5 vials
